L-asparaginase microbiana com potencial antioxidante

Resumo

A leucemia linfoide aguda é um tipo de câncer com rápida progressão no qual suas células necessitam da L-asparagina, uma vez que não possuem a asparagina convertase como as células saudáveis. A L-asparaginase excretada por bactérias vêm sendo utilizada como suporte no tratamento com quimioterápicos com grande sucesso. O presente estudo investigou o potencial de diversos microrganismos produzirem a enzima, utilizando a metodologia de planejamento fatorial obteve um aumento em 4,5 na produção da enzima, caracterizou parcialmente o complexo enzimático detectando um pHótimo correspondente 8,0 e temperaturaótima de 45°C, o complexo mostrou-se estável por 24h em uma ampla faixa de pH entre 3,0 a 8,0 manteve estável a temperaturas baixas de 35°C, apresentou atividade antioxidante frente aos radicais livres DPPH e ABTS apresentando IC50 de 3,879 e 3,76 mg/mL aos respectivos radicais livres avaliados. O presente estudo demonstrou Bacillus subtilis com potencial para a produção de L-ASNase com atividade antioxidante.
